Caracterización bioquímica y molecular de Trueperella (Arcanobacterium) pyogenes causante de abscesos en cerdos

Introducción: Trueperella (Arcanobacterium) pyogenes puede ser aislada de la piel, tracto respiratorio, sistema urogenital, gastrointestinal y membranas mucosas de animales domésticos. El microorganismo toma ventaja de las condiciones de estrés, infecciones primarias, innmunosupresión o trauma físico para invadir los tejidos epiteliales y causa infecciones purulentas. La principal limitante para el tratamiento, es la reincidencia de las infecciones tratadas con antibióticos y que en la actualidad no existe una vacuna eficiente en el mercado para su prevención. T. pyogenes ha sido aislada de cerdos con multiples abscesos en granjas del departamento del Tolima y este estudio fue diseñado para iniciar una caracterización microbiológica y molecular dirigida a la búsqueda de estrategias racionales para su prevención y control. Objetivo: Detectar proteínas antigénicas de una cepa de T. pyogenes (UT-Tp-12-001) aislada de abscesos en un cerdo reproductor raza Duroc – Jersey de una granja porcina en el departamento del Tolima, mediante las técnicas de SDS-PAGE, 2D-PAGE y Western blot Metodos: La identidad de la cepa de T. pyogenes (UT-Tp-12-001) fue confirmada mediante caracterización bioquímica y molecularmente a traves de la amplificación de un fragmento del gen PLO (piolisina) mediante PCR convecional. La cepa fue cultivada en agar sangre y masificada en agar tripticasa soya, para la extracción proteica mediante sonicación, los extractos fueron analizados por electroforesis en gel de poliacrilamida (SDS-PAGE), electroforesis de dos dimensiones (2D-PAGE) y tinción con azul de Coomasie. Las proteínas fueron analizadas por Western blot mediante transferencia a una membrana de nitrocelulosa e incubación con el anticuerpo primario policlonal, IgG Anti- T. pyogenes obtenido en conejo y el anticuerpo secundario, IgG de cabra anti-Conejo conjugado con la enzima fosfatasa alcalina. La reacción de los anticuerpos fue detectada mediante la adición del substrato 5-Bromo-4-Chloro-3-indolyl phosphate - nitro blue tetrazolium (BCIP-NBT). Resultados: El análisis mediante SDS-PAGE, reveló la presencia de proteinas con pesos moleculares de aproximadamente 18, 22, 27, 39, 50, 70 y 75 kDa. Las proteínas de T. pyogenes (UT-Tp-12-001) con pesos moleculares de aproximadamente 39, 50 y 75 kDa reaccionaron con el anticuerpo primario a diluciones de 1:5000 por Western blot. El análisis del perfil proteico de T. pyogenes (UT-Tp-12-001) a través de 2D-PAGE y tinción con azul de Coomasie usando el software PDQuestTM, evidenció 21 spots. La transferencia de dichas proteínas de T. pyogenes (UT-Tp-12-001) a una membrana de nitrocelulosa reveló 4 spots reactivos con el anticuerpo primario por Western blot. Estos spots de proteínas con un peso aproximado de 39 kDa, fueron detectados a diluciones del anticuerpo primario hasta de 1:2500. Conclusiones: Se describe la generación de un antisuero policlonal en conejo contra T. pyogenes (UT-Tp-12-001) que permitió identificar la antigenicidad de algunas proteinas de esta bacteria a través de Western blot. Estos resultados, permitieron establecer la presencia de una proteína con masa molecular aproximada de 50 kDa que retiene su antigenicidad a diluciones del antisuero hasta de 1:10.000. Esta proteína constituye una opción potencial para la elaboración de una herramienta de control y prevención de la enfermedad para región y el país. Introduction: Trueperella (Arcanobacterium) pyogenes can be isolated from the skin, respiratory tract, urogenital system, gastrointestinal and mucous membranes of domestic animals. The microorganism takes advantage of stress conditions, primary infections, innmunosuppression or physical trauma to invade the epithelial tissues and causes purulent infections. The main limitation for treatment is the recurrence of infections treated with antibiotics and that currently there is no efficient vaccine on the market for its prevention. T. pyogenes has been isolated from pigs with multiple abscesses in farms of the department of Tolima and this study was designed to initiate a microbiological and molecular characterization aimed at the search of rational strategies for its prevention and control. Objective: To detect antigenic proteins of a strain of T. pyogenes (UT-Tp-12-001) isolated from abscesses in a reproductive pig Duroc breed - Jersey of a swine farm in the department of Tolima, using the SDS-PAGE techniques, 2D-PAGE and Western blot Methods: The identity of the strain of T. pyogenes (UT-Tp-12-001) was confirmed by biochemical and molecular characterization through the amplification of a fragment of the PLO gene (piolisine) by conventional PCR. The strain was cultivated on blood agar and massified on trypticase soy agar, for protein extraction by sonication, the extracts were analyzed by polyacrylamide gel electrophoresis (SDS-PAGE), two-dimensional electrophoresis (2D-PAGE) and staining with Coomasie blue. The proteins were analyzed by Western blot by transfer to a nitrocellulose membrane and incubation with the polyclonal primary antibody, IgG Anti-T. pyogenes obtained in rabbit and the secondary antibody, goat anti-Rabbit IgG conjugated with the enzyme alkaline phosphatase. The reaction of the antibodies was detected by the addition of the substrate 5-Bromo-4-Chloro-3-indolyl phosphate-nitro blue tetrazolium (BCIP-NBT). Results: Analysis by SDS-PAGE revealed the presence of proteins with molecular weights of approximately 18, 22, 27, 39, 50, 70 and 75 kDa. The proteins of T. pyogenes (UT-Tp-12-001) with molecular weights of approximately 39, 50 and 75 kDa reacted with the primary antibody at dilutions of 1: 5000 by Western blot. The analysis of the protein profile of T. pyogenes (UT-Tp-12-001) through 2D-PAGE and staining with Coomasie blue using PDQuestTM software, evidenced 21 spots. The transfer of said T. pyogenes proteins (UT-Tp-12-001) to a nitrocellulose membrane revealed 4 spots reactive with the primary antibody by Western blot. These protein spots with an approximate weight of 39 kDa were detected at dilutions of the primary antibody up to 1: 2500. Conclusions: We describe the generation of a polyclonal antiserum in rabbit against T. pyogenes (UT-Tp-12-001) that allowed to identify the antigenicity of some proteins of this bacterium through Western blot. These results allowed us to establish the presence of a protein with an approximate molecular mass of 50 kDa that retains its antigenicity at dilutions of the antiserum up to 1: 10,000. This protein constitutes a potential option for the development of a disease control and prevention tool for the region and the country.
