Evaluación del efecto antisenescente del factor de crecimiento fibroblástico mediante la caracterización inmunofenotípica de un cultivo de células madre mesenquimales de gelatina de wharton

La capacidad de reparación de tejidos que presentan las CMM ha marcado importancia en los últimos años, en especial, CMM-gW, originadas del cordón umbilical, las cuales son una fuente útil en ingeniera de tejidos, a pesar de ser una fuente de fácil acceso, la cantidad necesaria para un trasplante exitoso es mayor a la adquirida en el aislamiento, por lo tanto se requiere de expansión a nivel in-vitro. Durante este proceso, la capacidad proliferativa de las CMM-gW se ve limitada provocando senescencia celular. Para atenuar este estado, se han propuesto diferentes factores de crecimiento entre los que se encuentra el FGF-2 el cual está presente en procesos de proliferación y migración celular, por lo tanto el objetivo de este proyecto fue evaluar el efecto del FGF-2 en el cultivo de CMM-gW, mediante la adición de dosis de 3,5 y 7,5 ng al medio de cultivo desde el pasaje 3, luego se evaluó la viabilidad celular, la senescencia del cultivo y se determinó la presencia de especies reactivas de oxígeno durante los pasajes 5 y 7, igualmente se realizó inmunofenotipificación. Se obtuvo que las dosis propuestas de FGF-2 promueven la proliferación celular y atenúan procesos como la senescencia y el estrés oxidativo, se resalta la actividad proliferativa de la dosis FGF-2 7,5ng, gracias a que demostró mitigar de manera significativa la senescencia celular. En conclusión las dosis propuestas de FGF-2 en este trabajo, contribuyen a disminuir moderadamente el proceso de senescencia celular, y reducir el estrés oxidativo. The CMM has been important in recent years, due to its proliferative characteristics and its differentiation to multiple lineages in especial CMM-gW, those derived from umbilical cord, which is a source of useful neurodegenerative diseases and other disorders, additionally these cells are characterized by their multipotentiality, capacity of adhesion to plastic surfaces and expression of positive surface antigens for CMM and absence of CMH markers. However, for a transplant is necessary an expansion process, in vitro unfortunately, the cells divisions decrease and it became senescent where their functionality and proliferative capacity reduced. Different factors have been selected among these FGF-2, which is present in different cell proliferation and migration processes. White the aim to evaluate the anti-senescent effect of FGF -2, were added doses of 3.5 ng and 7.5 ng to the culture medium since passage 3, and made it through inmunophenotypic characterization during passages 5 and 7. The cell viability and B-galactosidase positive cells, and were analyzed the levels of ROS in the cells. Results showed that two doses of FGF-2 facilitated cell proliferation and attenuate processes such as senescence and oxidative stress, in especial FGF-2 7.5 ng showed a better proliferative activity than other treatments. In conclusion, it was demonstrated that FGF-2 reduces the senescence levels of the CMM-gW and that the 7.5 ng dose was the one that showed the best results, the doses proposed in this project, reduce the process of cellular senescence, and reduce oxidative stress.
